It should show a speed up in playlists as the files will no longer need to be scanned. I don't use playlists so can't definitely say how much faster.
If your shared folders are being monitored then the database will update when files are added or removed.
If you move a file between shared folders it will treat it as a new file and re-scan it, and remove the old entry.
There is an option in the shared folder setup that allows you to set this.
